What is the difference between using PO13 to assign profiles to positions or using the Organizational Mgmt button on the User Tab of PFCF? Does RHPROL0 react differently? I'm trying to assign roles (or profiles) to positions, and things don't seem to be working, no matter which way I go. So, which way is the right way?

Either PFCG or PO13 do the same thing ( they establish an IT 1001 relationship) RHPROFL0 reacts identically to weither method as they both create the same record. Note ther eis a FROM-TO-DAte on the Info type.
You can see if RHPROFL0 will react correctly if you run tcode PPSS evaluation path O_TASKS.

As long as you choose/enter the correct values in to transaction PO13 there should be no problem.
You will however need to execute transaction PFUD in order to complete the user compare and finish the assignment to the position successfully. PFUD will also be required if you remove a role from a position.

PFUD or RHPROFL0 have an option to reconcile to the HR Org structure. Do note that IT 0105 Subtype 0001 has to be populated or nothing happens.
